Executive Summary

Accurate nucleic acid quantification is fundamental for the success of downstream genomic applications. This application note evaluates the performance of EzQuant Quantification Assay Kits across three leading fluorometric platforms: Blue-Ray Biotech EzCube, Thermo Fisher® Qubit™ 4, and DeNovix® DS-11 FX+.


By comparing accuracy across a range of concentrations and sample types, we demonstrate that EzQuant kits provide high sensitivity and cross-platform consistency for dsDNA, RNA, and ssDNA. Notably, the EzCube fluorometer delivers performance equivalent to the Qubit™ 4, providing industry-standard precision across essential laboratory workflows.

Introduction

The EzQuant Quantification Assay Kits utilize a highly specific fluorescent dye to bind precisely to target molecules at low concentrations, ensuring high sensitivity and minimal interference.


The suite includes specialized kits designed for specific detection requirements:

  • 1X dsDNA HS: Detects initial sample concentrations from 10 pg/µl to 100 ng/µl.
  • RNA HS: Detects total RNA initial sample concentrations from 0.25 to 100 ng/µl.
  • ssDNA: Detects ssDNA initial sample concentrations from 0.05 to 200 ng/µl.

Materials and Methods

To validate cross-platform consistency, five samples were tested for each kit:

  • Standard-Derived Samples: Three samples were prepared by diluting the kit's Standard 2.
  • Extracted Samples: Two samples were nucleic acid extracts obtained via silica membrane-based extraction to simulate real-world laboratory conditions.

All measurements were conducted on the EzCube, Thermo Fisher® Qubit™ 4, and DeNovix® DS-11 FX+. Results are reported in ng/µl.
